    Stability
    When stored as directed the product is stable for at least 6 months.
    *Store the entire kit under the conditions listed. Do not freeze Component A. Once the BacMam enhancer is reconstituted in DMSO, store the reconstituted product in DMSO at –20C for long-term storage. **Different amounts of BacMam enhancer (Component B) are included in B10019 and B10033 such that upon reconstitution in DMSO, each vial contains a 1000X stock solution of the product. Number of reactions: Sufficient material is supplied for 10 × 96 or 384 reactions (Cat. no B10019), or 100 × 96 or 384 reactions (Cat. no. B10033), based on the protocol below.
    Introduction
    The human ether-a-go-go related gene, or hERG (Kv11.1, KCNH2) encodes a voltage gated potassium channel known to repolarize cardiac muscle following an action potential. Blockage or perturbation of hERG activity in vivo leads to well described cardiac arrhythmias, and rigorous hERG screening and safety testing is required of all new drugs in preclinical development. To enable the study of hERG in a null background setting, we synthesized and packaged the full-length human clone of Kv11.1 into a baculovirus gene delivery/expression system. The use of baculovirus for transient gene expression in mammalian cells is referred to as BacMam technology (Figure 1). The BacMam-hERG reagent enables efficient delivery and transient expression in a wide range of mammalian cell types including human U-2 OS cells, and is used in conjunction with the novel FluxOR Thallium detection kits (Invitrogen Cat. nos. F10016, F10017) for functional screening of compound blocking activity at hERG. The FluxOR Thallium detection kit measures the activity of hERG and other potassium permeant channels with the report of thallium influx by a bright, fluorescent dye.
