摘要：本文基于已有的鸭血SOD提取工艺，对原有工艺进行优化。通过五因素四水平正交法探究热变性实验条件，得到热变性最佳温度为60摄氏度，最佳PH=7.0，热变性时间15min，同时在对Cu-Zn离子对热变实验影响的比较中发现，Cu-Zn离子共同作用能够最大程度的保持SOD活力。同时本文对于新工艺下提取的鸭血SOD进行了性质检测，使用邻苯三酚法测定SOD的活力，经过离子交换柱层析后的SOD活力为2150U/mg，电泳图像在16KDa有一条粗带，进一步证明SOD的成功提取。通过对比传统工艺和优化工艺的SOD的各项性质，可以证明优化后的提取工艺对于SOD的提取更加有效，为禽血SOD提取的工业化提供了实验指导，同时为大规模生产SOD提供了必要的实验支持。（5号，楷体GB2312）

__ This article is based on traditional duck blood SOD extract method, and optimize the process. By applying orthogonal method, we come to a conclusion that the optimum temperature for orthogonal is 60℃, and the optimum PH is 7.0, while 15min is best for orthogonality. Also, our research shows that Cu-Zn is needed for orthogonal method, when both of them are used in this method, it can best preserve SOD’s activity. Our research also detect the properties of the SOD extracting by the optimized method, we use pyrogallol method to test the activity of SOD, and the final product have an activity about 2150U/mg. electrophoresis image shows that a wide band accrues in 16KDa, which confirms the success of our research. By comparing the properties of SOD exacted by optimized technique and traditional method, we can find out that the optimized technique is more effective, thus to supply the process towards industrialization, and also offer the basic experimental data to the development of large-scale exacting of SOD.
